Anti-proliferation and Apoptosis Induced by Selective COX-2 Inhibitor in Human Esophageal Squamous Carcinoma Cells

چکیده
The human esophageal squamous cell carcinoma (ESCC) cell line (EC9706), which expresses Cyclooxygenase-2 (COX-2), was used to investigate the mechanism by which a new nonsteroidal anti-inflammatory drug (NSAID), NS398, inhibited proliferation and induced apoptosis in ESCC. The EC9706 cells were treated with various concentrations of selective COX-2 inhibitor, NS-398. Treatment with cox-2 selective inhibitor NS-398, significantly inhibited cell growth, induced apoptosis, and reduced the PGE2 production in a dose-dependent manner in human esophageal squamous cell carcinoma cells. Further investigation showed that NS-398 could significantly increase caspase-3 activity, and the caspase specific inhibitor AcDEVD-CHO could inhibit the increase of caspase-3 activity induced by NS-398. Our results indicated that NS-398 could Xu-vol5no1 3/10/05 6:34 PM Page 212

Cyclooxygenase-2 expression in human esophageal carcinoma.
On the basis of epidemiological observations that nonsteroidal antiinflammatory drugs reduce the risk of esophageal carcinoma, we studied the expression of cyclooxygenase-2 (COX-2) in esophageal squamous cell carcinomas (SCCs; n = 172) and in esophageal adenocarcinomas (ADCs; n = 27). Using immunohistochemistry, we observed COX-2 expression in 91% of the SCCs and in 78% of the ADCs. Western blo...
متن کاملInduction of Apoptosis by a Combination of 2-Deoxyglucose and Metformin in Esophageal Squamous Cell Carcinoma by Targeting Cancer Cell Metabolism
Background: Both mitochondrial dysfunction and aerobic glycolysis are signs of growing aggressive cancer. If altered metabolism of cancer cell is intended, using the glycolysis inhibitor (2-deoxyglucose (2DG)) would be a viable therapeutic method. The AMP-activated protein kinase (AMPK), as a metabolic sensor, could be activated with metformin and it can also launch a p53-dependent metabolic ch...
متن کاملEGFR activation results in enhanced expression of COX-2 and tumor growth through activation of β2-Adrenergic receptor in esophageal squamous cell carcinoma
We previously reported that epidermal growth factor (EGF)-induced esophageal cancer cell proliferation required transactivation of β-adrenoceptor. In the present study, we further investigated whether β2-adrenoceptor was involved in the modulation of cyclooxygenase-2 (COX-2) expression and cell proliferation by EGF in esophageal squamous cell carcinoma (ESCC). Human ESCC cell line KYSE-30 was t...

Objective(s): To analyze the potential influences of propofol on the oxidative stress of H2O2-induced human esophageal squamous cell carcinoma (ESCC) Eca109 cell through mediating the Wnt/β-catenin signaling pathway.Materials and Methods: Eca109 cells were classified into 5 groups: Control group, H2O2 group, Propofol + H2O2 group, Dkk1 (Dickkopf-1, Wnt/β-catenin pathway antagonist) + H2O2 group...
متن کاملThe Level of Mesenchymal-Epithelial Transition Autophosphorylation is Correlated with Esophageal Squamous Cell Carcinoma Migration
Background: The MET receptor is a critical member of cancer-associated RTKs and plays an important role in different biological activities, including differentiation, migration, and cell proliferation. Methods: In this study, novel MET inhibitors were introduced and applied on esophageal squamous carcinoma cell line KYSE-30, and the level of proliferation and migration, as well as the activated...
